I have a headache - should I cut off my head to fix it?

Anti-virus software...

Anti-spyware software...

Heck, you can break out rootkit revealer:

http://technet.microsoft.com/en-us/sysinternals/bb897445.aspx

But the only 100% effective thing to do is backup your data, wipe your machine, reload your operating system and programs and then put your data back. And then don't let anyone else touch your computer....

EDIT: did you start simple? Maybe she just has passwords to your email/facebook/myspace/whatever - change all your passwords to something random, put different ones on each site and see if that helps...

check task manager and google any suspicious processes.

use a friends PC to update your profiles/ myspace/blogs/etc. for a few weeks. see if she is still in the know.

Also you can't rule out the possibility of a snitch who is just reporting back to her. I've seen many a 'bro' dish dirt when they are allowed to reach into the cookie jar.
